چکیده
Abstract Reduced expression of glutamate decarboxylase 67 (GAD67), encoded by the Gad1 gene, is a consistent finding in postmortem brains patients with several psychiatric disorders, including schizophrenia, bipolar disorder and major depressive disorder. The dysfunction GAD67 brain implicated pathophysiology these disorders; however, neurobiological consequences mature are not fully understood because homozygous knockout lethal newborn mice. We hypothesized that tetracycline-controlled gene expression/suppression system could be applied to develop global knockdown mice would survive into adulthood. In addition, provide new insights impact dysfunction. Here, we developed tTA/STOP?tetO biallelic knock-in using STOP?tetO tTA mice, compared them +/+ level protein treated doxycycline (Dox) was decreased approximately 90%. GABA content also Dox-treated open-field test, exhibited hyper-locomotor activity duration spent center region. acoustic startle responses were impaired These results suggest reduction elicits emotional abnormalities will useful for elucidating mechanisms abnormalities, such as anxiety symptoms associated disorders.

Brain Glutamate Decarboxylase
Glutamate decarboxylase (glutamate 1-carboxylyase, EC 4.1.1.15, GAD)* IS the major, rate-limiting enzyme in brain for synthesizing gamma-aminobutyric acid (GABA). Total GAD activity in brain is 10-20 times greater than the observed rate of GABA synthesis (Collins, 1972; Matsui and Deguchi, 1977; Casu and Gale, 1981), indicating that GAD operates at only a fraction of its capacity. Although othe...

Mouse brain glutamate decarboxylase(s) was specifically titrated in vivo and in crude brain homogenates by a combination of gabaculine and [a-'H]acetylenic yaminobutyric acid. This specific titration is based on the differential spectra of action of these two mechanism-based enzyme inactivators. The specificity of the titration in vitro was demonstrated by showing that the time course of radioa...

Background: Gamma -aminobutyric acid (GABA), a non-protein amino acid acts as an inhibitory neurotransmitter in the central nervous system of mammalians. The glutamate decarboxylase (GAD) is responsible for the conversion of L-glutamate to GABA. The human brain has two isoforms of this enzyme, GAD65 and GAD67 that differ in molecular weight, amino acid sequence, antigenicity, cellular location ...
متن کاملPrevalence of autoantibodies to the 65- and 67-kD isoforms of glutamate decarboxylase in insulin-dependent diabetes mellitus.
We investigated the presence of autoantibodies to baculovirus-expressed human recombinant 65- and 67-kD isoforms of glutamate decarboxylase (GAD65 and GAD67) in insulin-dependent diabetes mellitus (IDDM). In the immunoprecipitation test using [35S]methionine-labeled GADs antibodies to GAD65 were detected in 13/15 (87%) islet cell antibody (ICA)-positive and in 1/35 (2.9%) ICA-negative first-deg...
متن کاملIdentification and molecular cloning of glutamate decarboxylase gene from Lactobacillus casei
Gamma-amino butyric acid (GABA) possesses several physiological functions such as neurotransmission, induction of hypotension, diuretic and tranquilizer effects. Production of GABA-enriched products by lactic acid bacteria has been a focus of different researches in recent years because of their safety and health-promoting specifities. In this study, glutamate decarboxylase (gad) gene of a loca...
